The series begins with the cold-blooded shooting of an unknown man as he tries to escape with a crying infant in his arms. When the shooting victim falls, mortally wounded, into a nearby river, the baby he is carrying floats away in a basin. The story continues many years later. The baby, Eli, is now a grown man. He is good-hearted and works hard at multiple jobs to help his family make ends meet. The four main characters meet for the first time at a special party onboard the luxury liner that the four main characters are together for the first time: Eli - working as a waiter; JB - the heir apparent to the shipping empire; Jackie - JB's girlfriend and the daughter of a wealthy and successful banker, who always tries to be the perfect daughter and perfect girlfriend,; and Celine - Jackie's cousin, who is also Jackie's exact opposite in attitude. Desperate circumstances due to his dad's illness and his desire to be with his wife abroad drive Eli's brother, Pong, to take part in an attempt to kidnap Jackie -- with disastrous consequences.

Sana Maulit Muli is about two young lovers who discover how true love can overpower fate and destiny. Small-town girl Jasmine Sta. Maria (Kim Chiu) and Travis Johnson (Gerald Anderson), the illegitimate son of an American soldier, both meet as stowaways in a ship sailing from Cebu. They promise to write each other as they were separated but this promise is intercepted as soon as Camille Soriano (Erich Gonzales) deceives Travis by becoming Poknat. In time, Travis' dad dies and their family leaves for the Philippines only to meet up with both Camille and consequently Jasmine. A rocky start between Jasmine and Travis turns into friendship then real love. Jasmine learns later on of Camille's deception and soon after learns that Camille is her younger sister. In order to make amends with her sister, Jasmine gives away everything for Camille's happiness leading to the break-up. Due to the taunting of Travis' brother, a twist ensues as Jasmine dies by a bus accident.

- GI Joe as Kokey - is an alien whose spaceship crashed on earth. Human-friendly and lovable, he will bring Bong to an adventure beyond the boy’s wildest dreams. His mission on earth is to find the missing stone that will lead to the discovery of Bong’s mysterious parentage.
- Joshua Cadelina as Bonifacio "Bong" Calundang/Gabriel Allegre - an orphan whose wild imaginations are often misunderstood. He tries to detach himself from others to avoid being rejected and for fear of losing a loved one again. Deep in his heart, he only wishes to belong to a family who will love and accept him as he is.

- Redford White as Nanding Calundang - the adoptive father of Bong. A Visayan businessman whose acumen in his field of expertise is truly remarkable. Although he quite lacks the need for competitiveness, his hard work still pays off. A very patient man, he dotes on his only daughter Anna.
- Eugene Domingo as Charisse - the archenemy of Trining. She is an ex-girlfriend of Nanding, who’s still bitter about their past. With a family of her own, she is determined to compete with Trining and show Nanding that she is a great loss in his life.
- Jojit Lorenzo as Marshall - Charisse’s husband and business partner of Nanding. He secretly resents his friend’s accomplishments as he continues to depend on him in their business ventures.
- Zanjoe Marudo as Abie - an aspiring pilot who will help Kokey build his spaceship. He immediately falls in love with Shane upon setting his eyes on her.

- Rica Peralejo as Sapphire Gomez / Bridget Ramirez - the eldest of the Jewel Sisters. When her parents died, Sapphire was separated from her sisters, and she later became Bridget Ramirez. She will eventually discover her true identity and will reunite with her two younger sisters. But along the way, Sapphire's lust for fame will pit her against Emerald and Ruby.
- Maja Salvador as Ruby Gomez / Kimberly "Kimmy" Bautista - the youngest of the Jewel Sisters. She was adopted by Berns Bautista who named her as Kimberly Bautista. But her dream of becoming a singer will lead her to Emerald and Sapphire. Ruby is obedient and quiet in nature, but her insecurities regarding her voice will be her greatest obstacle in life.She loves her sister and adopted Father very much.Poppy Burns and Emerald.
